Not exactly plug and play, but most of the Amiga OS used to come on a ROM chip, and a floppy disk was used just to tell the system to bring up the OS and also contain various utilities etc. Nice and responsive but not very easily upgradable (well, you could patch it using various programs if you had enough RAM).

That is roughly the same as a bootable cd/dvd version OS (Ubuntu, Knoppix). There are instructions on the web for making Ubuntu boot off of a flash drive.
